Abrus Precatorius Lectin Production Service

Abrus Precatorius Lectin Production Service

Abrus precatorius lectin (APA) is a natural lectin derived from jequirity beans. The seeds of jequirity beans contain nontoxic lectins and several toxic components that allow isolation using a variety of conventional techniques. APA has a variety of biological functions, including recognizing and binding specific glycosyl sequences, promoting cell aggregation, immunogenicity, and anti-tumor activity. CD BioGlyco provides professional Lectin Production Services, including Natural Lectin Production Service, Recombinant Lectin Production Service, and Lectin Purification Service.

The general extraction and isolation processes of APA are as follows. Firstly, soak the seeds of the jequirity bean in the water and stir it with a mixer. Then filter the suspension solution and collect it. Next, evaporate the filter fluid to remove most of the moisture. Finally, add an appropriate amount of solvents to the concentrated solution, so that the APA is precipitated in the solution.

Gel-filtration Chromatography-based Lectin Purification and Ion Exchange Chromatography-based Lectin Purification technologies are mainly used in the purification process. Before chromatographic purification, the precipitate is dissolved in an amount of water to form a crude extract solution of APA.

  • Ion-exchange chromatography: Use ion exchanges to exchange the APA crude extract solution to remove impurities. According to the types and concentrations of ion exchanges, the APA is further purified through multiple exchanges.
  • Gel-filtration chromatography: Using column layer analysis technology, the APA crude extract solution passes through the gel column. Separate the APA and other impurities according to the difference between the molecular size.

APA Identification Service at CD BioGlyco

  • Biological test: Use biochemistry methods (such as ELISA) to detect the concentration and activity of APA. The interaction between APA and specific antibodies are detected to evaluate the quality and purity of APA.
  • Molecular measuring detection: Use molecular sieve or gel electrophoresis technology to determine the molecular weight of the APA. The purified APA showes a single, clear band, and its molecular weight is consistent with the expected molecular weight.
  • Glycosyl group analysis: The type and proportion of the glycosyl group in APA are measured by gas chromatography or high-performance liquid chromatography.

Technology: The generation and comprehensive characterization of a panel of mAbs against abrin and APA

Journal: Toxins

Published: 2021

Results: The authors developed an efficient detection method to differentiate abrins from similar plant proteins. Initially, they produced precise monoclonal antibodies specifically targeting abrin and related lectins. Using these antibodies, they constructed two sandwich ELISAs capable of selectively identifying abrin-like lectins. Additionally, they designed a rapid on-site abrin-specific lateral flow assay. They demonstrated the versatility of the method in detecting abrins in intricate food and clinical samples.

Here are some of the results shown in this article:

Applications of APA Lectin

  • APA is used in western blot analysis to detect and identify specific sequences in proteins.
  • APA is used to study the processes of cell differentiation and apoptosis. It identifies and marks glycosyl expression on the cell surface through specific binding to glycosyl groups, thereby helping researchers understand the growth and death mechanisms of cells.
  • APA is used for virus detection and identification. It identifies and labels viral particles by binding to sugar groups on the virus surface, thereby helping researchers determine the type and characteristics of the virus.
