Background
Glycosylation is a common post-translational modification (PTM) of proteins. This feature is caused by changes in sequence and chain length, anomeric configuration (α or β), connection positions, and branch sites. The covalent modification of a single glycan residue through sulfation, phosphorylation, acetylation or methylation adds further structural variation to the carbohydrate chain. Therefore, the structural diversity available in glycan chains far exceeds the complexity of obtaining amino acids in polypeptides. Two of the most abundant forms of protein glycosylation are N- and O-linked glycosylation. N-linked glycans are usually linked to asparagine (Asn) via GlcNAc. Mucin-type O-linked glycans are linked to Ser or Thr via GalNAc, but other O-glycans can be linked to Ser/Thr via GlcNAc, fucose, glucose, or mannose.
Glycan-modified Microarrays at CD BioGlyco

The glycocalyx of human cells is different from that of many other mammals in that it lacks the sialic acid N-hydroxymethylneuraminic acid (Neu5Gc), and the abundance of its precursor hydroxylated form N-acetylneuraminic acid (Neu5Ac) is increased. Neu5Gc is recognized as a foreign body by the human body's fluid immune system. This is especially important when using non-human cells to produce antibodies. These modifications have a huge impact on the structure and function of glycoproteins. Microarray tools have far-reaching significance for rapid analysis and detection of these modifications and analysis of glycoproteins.
