  • How does IEC work?

IEC is a widely used chromatographic method for lectin purification. Its stationary phase is a chemically bonded ion exchanger or ion exchange resin. This method utilizes the difference in selectivity coefficients or ion exchange capacity of the components being separated. Different biomolecules have different charges and therefore they have different binding capacities to ion exchangers. When eluted, these substances are eluted off in a different order, thus achieving separation.

  • We combine affinity chromatography on mannose-sepharose with IEC to obtain a highly pure preparation oflectins.
  • Some lectins are purified by a combination of ammonium sulfate precipitation, dialysis, IEC, and gel filtration chromatography.

Results: In this study, cation exchange chromatography coupled with gel filtration chromatography was used for the isolation and purification of lamprey (Lampetra japonica) serum. Elution fractions were collected based on hemagglutination activity, resulting in a polymeric lectin (105-kDa). The purification method was tested to be efficient and yielded a single fraction.

Applications
