  • Firstly, we use monosaccharides or disaccharides as glycosylated donors and prepare tri-, tetra-, and other oligosaccharides by glycosylation. In this process, we provide chemical modifications such as selective sulfation, sulfonation, acetylation, or hydrogenation to extend the backbone structure of the oligosaccharides.
  • Then, we utilize glycosyltransferases to modify the oligosaccharides prepared in the previous step to further extend the structure and type of oligosaccharides and synthesize oligosaccharide fragments efficiently.
  • Next, oligosaccharide units with different main chain structures or different branched structures are assembled efficiently, respectively. Among them, we provide chemical reagents to regulate and realize the construction of glycosidic bonds with high stereoselectivity.
  • Finally, we utilize an orthogonal one-pot glycosidation reaction to complete the assembly of the target oligosaccharide library.

Throughout the process, we offer nuclear magnetic resonance (NMR), high-performance liquid chromatography (HPLC), and electrophoresis to analyze the structure of the oligosaccharides. We also offer weak anion exchange column chromatography to remove impurities and unreacted substances.

Technology: Modular disaccharide assembly of tetrasaccharides with selective protection

Results: The authors describe a synthetic method for the construction of heparan sulfates (HS) oligosaccharide libraries. The structure is based on the use of modular disaccharides to form selectively protected tetrasaccharides. These tetrasaccharides have been subjected to selective chemical modification such as regioselective O- and N-sulfation and selective desulfation and enzymatic modifications. A total of 12 different HS derivatives were obtained. A library of 47 HS oligosaccharides has been developed and the resulting compounds have been synthesized to create HS oligosaccharide microarray, using tetrasaccharide backbone compositions with various compositions. The ligand requirements of many HS-binding proteins, such as fibroblast growth factor and chemokines, were examined using this array.
