Physiological Effects of Lactitol

The active ingredient of lactitol is lactitol monohydrate, which is a monosaccharide derivative formed by linking galactose and sorbitol through glycosidic bonds. The chemical name of lactitol is 4-O-β-D-galactopyranosyl-D-glucitol. Lactitol is an oral osmotic laxative, which is rarely absorbed systemically and is not absorbed in the intestine. It is metabolized by colonic microorganisms into D-galactose and D-sorbitol, which are fermented into organic acids, containing lactic acid, formic acid, propionic, butyric, and acetic acids. Through osmosis, extra fluid is transported to the colon, thereby promoting intestinal peristalsis, and acting as a laxative. Moreover, studies have shown that supplementing lactitol also improves the composition of fecal flora and increases the content of Bifidobacteria.

Lactitol Analysis Service at CD BioGlyco

Analysis of lactitol is critical as it is widely applied in the food and pharmaceutical industries. CD BioGlyco provides lactitol analysis services for our clients worldwide by the following methods.

  • High-performance Liquid Chromatography (HPLC)
HPLC is a common analysis method, which is used to determine the content and purity of lactitol. It separates the lactitol in the sample from other components under pressure and carries out quantitative analysis through the detector.
    • Evaporative light scattering detector (ELSD)
    • Pulsed amperometric detector (PAD)
    • Refractive index detector (RID)
  • Gas Chromatography (GC)
It is usually used in conjunction with pretreatment steps, for example, derivatization reactions, to convert lactitol into compounds that are separated and detected on a GC column.
  • High-performance Anion-exchange Chromatography (HPAEC)
It is usually used in conjunction with a PAD detector to test the content of lactitol.
  • Chromatography-Mass Spectrometry (MS)

Paper Title: Determination of maltitol, isomaltitol, and lactitol by high-pH inion-exchange chromatography with pulsed amperometric detection

Technology: HPAEC-PAD

Journal: Journal of agricultural and food chemistry

IF: 5.895

Published: 1999

Results: HPAEC-PAD is a very powerful tool for the test of sugars and sugar alcohols. The authors developed an optimized HPAEC−PAD method using a thin-film anion-exchange column with a low ion-exchange capacity to quickly and efficiently separate sugar alcohols, containing lactose, present in many commercially available nutraceuticals using a highly basic mobile phase. Lactitol and other sugar alcohols were rapidly separated under optimized conditions without the need for strong base addition to the eluent post-column and without column regeneration between runs. This way is simple, sensitive, and reproducible.

Applications

  • Food production: Lactitol is widely applied in food processing as a sweetener. Lactitol analysis is used to evaluate the content and purity of lactitol in food to ensure that the product meets regulatory standards.
  • Pharmaceutical field: Lactitol analysis is used to determine the content, purity, and stability, which ensures drug quality and safety.
  • Study and development: Through the accurate test of lactitol content, its functionality, sweetness characteristics, and interaction with other ingredients are evaluated to provide support for product development.
