Biological Activity of Glucosamine

Glucosamine is a compound obtained by replacing a hydroxyl group in the glucose molecule with an amino group. It is widely found in animal cartilage and crustacean shells. Glucosamine can promote the synthesis of cartilage and inhibit its breakdown, with the ability to combat the damage done by inflammation and stimulate the growth of cartilage cells. Glucosamine can also protect lipid macromolecules from oxidative damage by hydroxyl radicals, showing strong antioxidant activity. In addition, it has the function of stimulating the immune response and enhancing the body's immunity.

  • Preparation of solutions
    • Solid sample: The solid is accurately weighed, fully dissolved in trifluoroacetic acid (TFA) solution, and then blow-dried with nitrogen gas. Repeated cleaning with methanol is performed several times. Finally, the sample is dissolved in sterile water and put into a clean chromatographic bottle.
    • Liquid sample: The liquid is concentrated or blow-dried with nitrogen gas into a solid. The subsequent steps are consistent with the preparation of the solid sample.
    • Standard solution: The glucosamine powder is accurately weighed and fully dissolved with sterile water to prepare a control solution.
  • Instrumental analysis: Ion chromatography system and liquid chromatography column are used. The injection volume is 5 μL. Mobile phase A is H2O, and the NaOH solution works as mobile phase B. The glucosamine component is detected and analyzed by the electrochemical detector.
  • Qualitative and quantitative analysis: By high-performance liquid chromatography (HPLC) separation, the glucosamine component is qualitatively analyzed based on the retention time of the standard solution and components in the sample. The concentration-peak area curve is drawn by testing standard solution, and the concentration of glucosamine in the sample is calculated according to the peak area.
