Synthetic Glycan Microarray Assay

Synthetic Glycan Microarray Assay

Unlocking the Power of Glycans, One Spot at a Time

To help researchers investigate the interactions of a variety of glycan with biomolecules, such as proteins, antibodies, etc., glycan microarray technology is used. Currently, CD BioGlyco has constructed a powerful and comprehensive Glycan Display Platform that provides several types of synthetic Glycan Microarray Assay services including N-Glycan Microarray as well as O-glycan microarrays and other Glycan Microarray. The technologies and services we offer are shown below.

Synthetic Glycan Microarray Assay Service at CD BioGlyco

  • We offer to construct the N- or O-glycan microarray.
    • We immobilize N- or O-glycans on slides by using them for covalent printing.
    • The slides are then incubated with compounds capable of binding N- or O-glycans. The binding of the compounds to the N- or O-glycans is detected by direct or indirect fluorescence methods (the image of a fluorescent spot whether or not it binds to the deposited N- or O-glycans). In this way, those recognized glycan ''dots'' are identified and can be visualized by direct or indirect fluorescent labeling.
    • Finally, the fluorescence intensity is measured and plotted on a microarray to obtain an N- or O-glycan microarray.
  • The specific N- or O-glycan microarrays we offer include, but are not limited to:
    • Conventional glycan microarrays
    • Microbial glycan microarrays

We offer N- or O-glycan microarrays with over 90 different glycan structures, and each array contains multiple identical subarrays, allowing for simultaneous analysis of multiple samples. Sample types include, but are not limited to, proteins, cells, antibodies, cell lysates, viruses, and more.

Results: The authors develop a method for examining custom glycan microarrays for glycan and glycoprotein binding specificity. The binding-ready synthetic glycans are partially and covalently attached to the surface of the microarrays via nucleophilic junctions. Incubation of the microarrays with glycoproteins and quantitative detection of the binding process by fluorescent signaling. Such methods are quite easily adapted to a variety of goals spanning from basic research to bariatric biomedical applications.

Applications

  • Synthetic glycan microarray assay can be used to assess glycan interactions and binding specificity between glycans and proteins.
  • Synthetic glycan microarray assay can be used to evaluate the binding specificity of glycans to antibodies.
  • Synthetic glycan microarray assay can be used to research viral, bacterial, vesicular, and cellular interactions with glycans.
