Results: This article describes a method for the determination of streptomycin in apples. The method utilized LC-MS/MS techniques and was validated according to SANCO/3131/2007. First, sample preparation was carried out by phosphate buffer extraction and pH change, followed by a clean-up using polymeric phase SPE. The LC-MS/MS analysis employed a hydrophilic exchange column to separate the analytes present in the samples, while a triple quadrupole MS was utilized to measure the conversion of substances in positive ESI mode. To quantify streptomycin, a calibration curve using matrix standards with a linear range spanning from 1.0 to 20 μg/kg was established, alongside an internal standard dihydrostreptomycin at a concentration of 10 μg/kg. The performance of the method was verified by calculating the recoveries (101-105%) and the relative standard deviations (RSDs, 4.1-11.4%) of six replicate samples. The quantification limit of this method was determined to be 2 μg/kg.
