Introduction of Next-generation Sequencing (NGS)

The rapid update of RNA-seq technology has accelerated transcriptome analyses in new times with higher efficiency and lower cost. Unlike the first-generation sequencing methods based on Sanger sequencing, which used fluorescently labeled or radioactive dNTPs, the researchers used a recently discovered luminescent method named pyrosequencing for DNA sequencing. It laid the foundation for the emergence of NGS. NGS uses "sequencing by synthesis" technology, making it possible to sequence hundreds of thousands of nucleic acid molecules at once and analyze the transcriptome and genome of a species in detail. Therefore, it is also called high-throughput sequencing, which can easily detect unknown gene expression sequences, with many advantages such as fast speed, low cost, and high precision.
