Background

As the product of protein glycosylation modification, glycoprotein is one of the common forms of protein in organisms. It plays an important role in various life activities. The abnormal expression of glycoproteins is closely related to the occurrence and development of a variety of diseases. Glycomics analysis of functional glycoproteins is of great significance to deeply reveal the structural changes of glycoprotein oligosaccharide chains and their relationship with the corresponding functions.

Chemical methods have been used to release O- and N-linked oligosaccharides. Several chemical release techniques have been applied for a long time, of which the most commonly used methods are hydrazinolysis and alkali/reducing conditions (β elimination). The release of N-linked glycans using anhydrous hydrazine was mainly developed by Kobata group and has been used by many groups for the analysis of a large number of glycoproteins. Moreover, this technique can also be modified for the release of O-glycan structure. At present, the new methods described in numerous reports overcome the shortcomings of traditional methods and can obtain more abundant glycan structure and function information, which has important application value in glycomics research.

Release Strategies

CD BioGlyco has established a series of simple, economical, and rapid chemical methods for the nonspecific release of glycan chains. The services we can provide include but are not limited to:

  • In terms of O-glycans, we established a new method of universal and reliable non-reductive release and simultaneous derivatization labeling and a new method of relative quantitative analysis based on stable isotope labeling. The methods not only simplify the preparation process of glycan chains sample, but also avoid the interference of by-products, and greatly improve the glycan release yield, analysis efficiency, and analysis reliability.
  • In terms of N-glycans, we have established methods of non-reducing release, reducing release, simultaneous release, labeling, and corresponding new methods of qualitative and quantitative analysis. These methods have the advantages of low cost, simple operation, and high yield of glycans, which can not only realize the large-scale preparation of N-glycans but also provide a material foundation for the biological activity research of natural N-glycans.
