Workflow

Results: This graduate student generated multiple different polypeptides and linked several polysaccharides (the primary structural components of bacterial cell walls) to produce various PGN fragments. Ultimately, using a novel chip synthesis technology, a microarray containing hundreds of PGN fragments from Gram-positive and Gram-negative bacterial variants was generated. Researchers validated the array and its epitopes using four monoclonal antibodies and analyzed plasma samples from epidermolysis bullosa (EB) patients using these microarrays to validate the antibody profile. The results showed that surface functionalization of PGN plays a key role in IgG responses in patients. This study demonstrates the potential application of PGN microarrays in identifying specific antibody profiles targeting the human microbiome, particularly in intestinal diseases.
