Properties and Biosynthesis of PI

PI is an essential acidic phospholipid, which exists in the membrane of mammalian cells and chromatin. PI is mainly composed of two parts: 1,2-diacylglycerol phosphate and inositol. The inositol ring of PI can be phosphorylated at three positions alone or in combination to produce seven derivatives. PI plays important roles in cell morphology, metabolic regulation, signal transduction, and various physiological functions. In addition, the PI can be phosphorylated to form PI phosphate (PI-4-P), PI diphosphate (PIP2), and PI triphosphate (PIP3). All PI-based lipids are known as inositides, sometimes phosphoinositol.

The de novo biosynthesis of PI starts from the precursors 3-phosphoglycerol or dihydroxyacetone phosphate and the process only occurs in the endoplasmic reticulum (ER). These molecules undergo two sets of acylation under the action of acyltransferase to form lysophosphatidic acid (LPA) and phosphatidic acid (PA), respectively. The synthesis of PI in the laboratory is catalyzed by phosphatidylinositol synthetase, involving CDP-diacylglycerol and L-inositol.
