Cell-based Glycoprotein Array

Cell-based Glycoprotein Array

Your Best Partner for Cell-based Glycoprotein Array

Glycoproteins are an important class of glycoconjugates. The various biological functions of glycans are also exerted mainly through their participation in various important biological processes with the protein backbones. The composition of oligosaccharide structures in glycoproteins produces various glycosylated structures depending on their cell type, organization, and species. Glycosylation also affects glycoprotein functions, including but not limited to immunogenicity, tissue targeting, and pharmacokinetics. CD BioGlyco dissects intermolecular interactions and the biological functions of glycoproteins through Cell-based Arrays and provides a wide range of biological analysis services to our clients.

  • Construct cell-based arrays displaying glycoprotein
  • CD BioGlyco constructs glycoprotein-producing cell lines by transfecting genes in parental cell lines under selection pressure and repeating several rounds of clonal selection. We offer two main types of cell lines: Chinese hamster ovary (CHO) and human embryonic kidney 293 (HEK293) cells. The selection of cell lines with the ability to produce high yields of glycoproteins with adequate glycosylation patterns is essential for efficient glycoprotein acquisition. our researchers recommend the best solution based on the characteristics of your project.
  • With the support of nuclease genetic engineering, our talented team engineers mammalian cells by combinatorial knockout (KO) and knockin (KI) of glycosylated genes. To explore glycoprotein-dependent biological interactions by displaying unique features of glycoproteins on the cell surface. Furthermore, based on stable and robust genetic engineering, we have developed isogenic HEK293 cell libraries with different glycoproteins for displaying glycoproteins with different glycosylation abilities.
  • High throughput screening service

At CD BioGlyco, we provide the specific binding assay service and substrate tolerance assay service for cell-based glycoprotein arrays. Moreover, we provide array screening services in identifying high-affinity ligands. Our analysts characterize the protein-binding properties to further analyze the interaction relationship between cell-based glycoprotein arrays and viruses.

Paper Title: Display of the human mucinome with defined O-glycans by gene engineered cells

Technology: Genetic engineering, Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), Mass spectrometry, Flow cytometry

Journal: Nature Communications

Published: 2021

IF: 16.6

Results: In this study, cell-based mucin displays were constructed from HEK293 isogenic cells and bound to three adhesins (HsaBR, GSPBR, and NCT10712BR), respectively. All three adhesins were found to bind tightly to different O-glycan structures and different subpopulations of mucin tandem repeats (TRs). Analysis of the binding properties of the adhesion proteins identified specific O-glycan structures and screened for mucin TR sequences. The high-intensity mucins TRs display information indicates the mucin display platform for the discovery and dissection of interactions with O-glycans on mucins.

Applications of Cell-based Glycoprotein Array

  • Cell-based glycoprotein array screening for the toxic effects of natural compounds on cancer cell surface proteins can be used for the development of antitumor agents and glycoprotein inhibitors.
  • A cell-based glycoprotein array is an important tool for exploring the glycan and protein interactome.
  • Cell-based glycoprotein arrays are an effective tool for exploring and resolving mucin-degrading enzymes, such as proteases secreted by pathogenic glycoproteases of the C1 esterase inhibitor (StcE).
  • Cell-based mucin arrays provide an extremely important display platform for the discovery and high-throughput analysis of the biomolecular properties of natural mucins and other glycoproteins with mucin domains.
  • Cell-based glycoprotein array is an important target in the field of studying recombinant proteins and the microbiome and is an effective tool for analyzing and screening the biological functional properties and substrate binding of glycoproteins.
