  • Identification
    • The sample to be examined is placed under the microscope after adding an appropriate amount of iodine test solution. Most of the cells of guar gum are round, pear-shaped, or polyhedral, and their intracellular material is yellow to brown.
    • Add an appropriate amount of water to the sample to be tested and shake vigorously, if a gel or semi-gel appears, the sample is proven to be guar gum.
    • A moderate amount of sodium tetraborate solution is added to an aqueous solution of the sample, if this sample is guar gum, the solution will appear as a translucent elastic gel.
  • Content analysis
    High-performance liquid chromatography (HPLC) is used to determine the amount of guar gum in samples. In this, water is used as the mobile phase, with a cation exchange resin serving as the filler.
  • Insoluble substance analysis in acid
    Water and sulphuric acid are added to the guar gum and then heated in a water bath. After a while, the water is removed and it is left to dry at 105°C. The residue remaining after drying should not exceed 7.0 percent.
  • Starch analysis
    Add iodine test solution to the aqueous solution of guar gum, if a blue color appears, it means that guar gum contains impurity starch.
  • Protein analysis
    We test for the presence of protein in guar gum using a nitrogen assay. Notably, the protein content of guar gum should not exceed 10.0 percent.
  • Borate analysis
    The guar gum is dissolved in water and a hydrochloric acid solution is added. Use turmeric test paper for testing: if the guar gum solution appears brownish red and the color of the test paper deepens after drying, or if it turns dark green after being wetted with the ammonia test solution, it proves that there are impurity borates in the guar gum.
  • Loss on drying
    The guar gum is dried at 105°C for 5 hours with a weight loss of not more than 12.0%.
  • Microbiological limit analysis
    We use the flat dish method, membrane filtration method, and the most-probable-number method (MPN method) to detect the enumeration of thermophilic bacteria and fungi in guar gum grown under aerobic conditions.

Results: In this paper, the authors utilized an in-house constructed THz-time domain spectroscopy (THz-TDS) system to intensively research the THz absorption properties of guar gum in different environments (concentration, temperature, and electric field exposure time), leading to the development of an efficient technique for identifying guar gum colloids. After a series of experiments, the authors found that the intensity of THz transmission of guar gum decreases with increasing temperature, and a higher concentration of guar gum solution corresponds to lower THz transmission intensity, resulting in stronger terahertz absorption. Moreover, the THz absorption intensity of the guar gum solution showed a decreasing trend with the increase of the exposure time to the electric field.

  • What are the structural characteristics of guar gum?
    Guar gum is a non-ionic polysaccharide as far as molecular structure is concerned, with polymannose as the molecular backbone and D-mannopyranose units linked by β(1→4) glycosidic bonds. D-Galactopyranose, on the other hand, is attached to the polymannose backbone with an α (1→6) bond. The molar ratio of mannose to galactose units in guar gum is 2:1, i.e., every other mannose unit is connected to a galactose branch. The most important feature and advantage of the guar gum molecule are that it is very similar in structure to cellulose, and this similarity gives it a strong affinity for cellulose, called substantivity.
