Results: The authors had developed a customizable flux-conditioned FC-E8 medium that permitted stem cell cultivation with stable isotopes for metabolic tracking, alongside a specific liquid chromatography-mass spectrometry (LC-MS/MS) technique that focused on metabolic pathways integrating in nucleotide sugar biosynthesis. Through the utilization of 13C6-glucose supplementation, they were able to track carbon integration into nucleotide sugars over time, both directly via glucose and indirectly through alternate pathways like glycolysis and pentose phosphate pathways, within induced pluripotent stem cells and embryonic stem cells.
