GS is an organic substance with the chemical formula C60H125O22N15·H2SO4. It has the appearance of a white or off-white crystalline powder and dissolves easily in water. In biology and medicine, GS has a favorable inhibitory effect on most bacteria.   • Specific rotation
    We accurately weigh a sample of GS and dissolve and dilute it with water. Next, the specific rotation of GS is tested using a spinometer. GS should have a specific rotation of +107° to +121°.
  • Solubility
    GS is soluble in water and insoluble in ethanol, acetone, or ether.
  • Identification
    • Thin layer chromatography (TLC): We will dissolve the sample to be tested and GS standard in water and make it into separate solutions. Next, both solutions are unfolded on the same thin layer plate using the same unfolding agent. Finally, we compare the spots of the sample and the standard in iodine vapor. If the number, location, and color of the spots match, the sample is proven to be GS.
    • Infrared spectroscopy (IR): Infrared light absorption spectra of the control sample and the standard of GS are compared, if the spectra are the same, it proves that the sample is GS.
  • Acidity
    The pH of an aqueous solution of GS is determined directly using an acidimeter. The pH of GS should be 4.0-6.0.
  • Sulfate
    GS is weighed precisely and dissolved in water. We titrate it using barium chloride solution as titrant and phthalide violet solution as indicator solution. Finally, the amount of sulfate in GS is calculated based on the color change and the amount of titrant used.
  • Quantitative analysis
    We use high-performance liquid chromatography (HPLC) to determine the content of GS. The content of GS is calculated using the area of the peak of GS in the chromatogram obtained.
  • Moisture
    We determine the moisture content of GS using a combination of a moisture meter and an electrochemical method.
  • Bacterial endotoxins
    We offer gel and photometric methods for the determination of endotoxin in GS. These two methods mainly utilize the agglutination phenomenon and turbidity change during the reaction of horseshoe crab reagent with endotoxin to determine the amount of endotoxin.

Results: The authors crosslinked two alginate gels of different compositions with Ca2+ and exposed them to GS at different times and concentrations. The authors then found that GS was effective in improving the physicochemical properties of the alginate network by determining the mechanical properties of the formed complexes, NMR spectra, and pH, among others, and that the extent of these changes was related to the structural composition of the polysaccharides.
