  • Process of natural oligosaccharide library construction at CD BioGlyco
    We isolate and characterize naturally occurring oligosaccharides from different sources as a way to construct natural oligosaccharide libraries. The process of construction is as follows.
    • Source Selection
      We select the source from which you want to isolate the oligosaccharides. Natural oligosaccharides are derived from a variety of biological materials, including plants, microorganisms, animals, algae, etc.
    • Material Pre-treatment
      Pre-treatment is performed to facilitate the release of the glycan. Biological materials (cells, tissues, etc.) are impregnated, ground, or homogenized.
    • Extraction of Oligosaccharides
      We extract the oligosaccharides using appropriate solvents. We use different solvents such as water, salt solutions, etc. for extraction. The choice of solvent depends on the solubility and stability of the target oligosaccharide. This process is combined with enzymatic hydrolysis as a way to obtain structurally diverse oligosaccharides.
    • Separation and Purification
      We separate and purify the extracted oligosaccharides by filtration, size exclusion chromatography (SEC), and other methods.
    • Analysis and Characterization
      The structure, composition, and modification of the isolated oligosaccharides are characterized by liquid chromatography (LC), mass spectrometry (MS), nuclear magnetic resonance (NMR), and other methods.

Results: In this study, a Miscanthus sinensis grass glucuronoarabinoxylan (GAX) stem and leaf oligosaccharides reference library was constructed by hydrolyzing endo-β-xylanases GH10 and GH11. The library is expressed in glucose units. Analysis of the GAX structures of several species using this library shows that three main GAX sugar side chains are used to decorate xylan molecules. This library provides a very efficient method for xylan structure studies.
