  • Optimization of wash using colorimetric reporter phage

Our researchers use optimization of wash using colorimetric reporter phage to test four different phage clones simultaneously without sequencing. Meanwhile, we provide testing of the input and output quantities of phages. This service makes the project go more smoothly, and it is very convenient.

  • Quality control by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS): Our investigators perform a detailed analysis of the glycosylation degree of phage samples by MALDI-TOF-MS and provide coupling analysis services for different glycan structures.
  • Quality control by deep sequencing: Through deep sequencing analysis, we provide clients with molecular binding properties between glycans and glycoproteins. The service report contains key binding information in vitro or in vivo regarding glycoprotein interactions of glycan and cellular reality.

Results: In this study, constructed LiGAs were acylated using dibenzazo-cyclooctyl-N-hydroxysuccinimide ester (DBCO-NHS), and their linkage was detected by MALDI-TOF-MS. The results indicate that the degree of coupling interferes with binding to antibodies or glycoproteins, presumably due to the spacing of pVIII on the phage surface. In addition, the coupling of different glycan structures was determined by MALDI analysis (shown below). It also provided information on the position of the amide bond mainly on the N-terminal amine.
