What Is Lactose?

Lactose, also known as β-D-galactopyranosyl-(1→4)-D-glucose, is a reducing disaccharide formed by the condensation of galactose and glucose, and both α-anomeric and β-anomeric are present. It is the major carbohydrate in most mammalian milk and makes an important contribution to the nutritional needs of newborns. Due to its physiological and functional properties, lactose has been widely used in the food and pharmaceutical industries. For example, lactose can be used as a free-flowing or agglomerating agent to enhance food flavors or as a diluent for colors, flavors, or enzymes. In the pharmaceutical industry, lactose is widely used as a drug carrier because it is safe, highly refined, and not sweet.

Lactose Metabolic Pathway

After lactose is internalized into cells, it is metabolized through Leloir and tagatose-6-phosphate pathways. In the Leloir pathway, lactose is hydrolyzed into glucose and β-galactose by β-galactosidase. β-Galactose is then transformed into glucose-1-phosphate via galactose mutarotase (GALM), galactose kinase (GALK), galactose-1-phosphate uridylyltransferase (GALT), and uridine diphosphate-galactose 4'-epimerase (GALE). In the tagatose-6-phosphate pathway, lactose-6-phosphate is hydrolyzed to glucose and galactose-6-phosphate by phospho-β-galactosidase (lacG). Galactose-6-phosphate is then transformed into glyceraldehyde-3-phosphate (GPDH) and dihydroxyacetone phosphate (DHAP) via galactose-6-phosphate isomerase (lacAB), tagatose-6-phosphate kinase (lacC) and tagatose-1,6-diphosphate aldolase (lacD).
