  • Extraction and purification services

AMA is a monocotyledonous plant lectin, which is extracted and isolated from the tuber of calla lanceolate. We crush the taro tuber, dissolve it in saline, and obtain its crude product through stirring, centrifugation, filtration, and dialysis.

For the purification of AMA, affinity purification of desialic acid fetal globulin by fixed column chromatography and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) are excellent ways.

  • Property testing services
    • Structural composition
      The protein composition of the lectin is tested by SDS-PAGE and gel filtration, and the subunit composition of the lectin is tested by sequencing the N-terminus of the protein in combination with the molecular cloning of the lectin.
    • Molecular weight
      The total molecular weight of the lectin is identified using polyacrylamide gel electrophoresis, and the molecular weight of its constituent subunits is identified using SDS-PAGE.
    • Cross-reactivity
      The cross-reactivity of purified lectin is verified by protein blotting using an AMA antibody.
    • Target receptor proteins
      The primary receptor protein for this lectin is detected from brush border membrane vesicle proteins of the intestine using ligand binding. Upon testing, α-d-mannose is found to have a blocking effect on this binding of AMA to the BBMV receptor.
    • Coagulation activity assay
      A microplate is taken, and the same concentration of AMA protein is added to each of the four wells, after which different concentrations of rabbit erythrocyte suspensions are injected respectively, and the plate is placed on an oscillator at a medium speed so that the coagulation effect is observed by the naked eye or under a microscope.

Publication

Technology: Models of neutrophil migration in vivo, neutrophil chemotaxis in vitro, and macrophage cultures were used to study the lectin inflammatory activity.

Results: The AMA exhibits different specificities for N-acetyl lactose amide type and glycan oligosaccharide-mannoside type. The authors researched the action of this lectin on cells of the immune system. The inflammatory activity of the lectin was also researched using models of neutrophil migration in vivo, neutrophil chemotaxis in vitro, and macrophage culture. When applied to the peritoneal cavity of rats, AMA induced significant and dose-dependent neutrophilic leukocyte migration. Secondly, pretreatment with mercaptoacetate increased the number of peritoneal macrophages, thereby enhancing AMA-induced neutrophil migration. Reducing the number of peritoneal mast cells, however, did not alter AMA-induced neutrophil migration. Further experiments revealed that lectins induced neutrophil chemotaxis. In conclusion, all data suggested that the lectin had pro-inflammatory activity and induced neutrophil migration.
