B. bubalis lectin (BBL) is a soluble β-galactoside binding lectin with the highest affinity for lactose and other glycol groups in the glycosidic form. With a molecular weight of 14.2 kDa, the neutral sugar content of the soluble lectin is estimated to be 3.3%. BBL Extraction and Purification Service at CD BioGlyco

CD BioGlyco has many years of successful extraction and separation experience. Selection of fresh nerve tissue, wash, remove meninges, and cut into small pieces. The tissues are homogenized in phosphate compound buffer and centrifuged to obtain supernatant. A mixture of ammonium sulfate precipitation is added to the supernatant, followed by gel filtration to collect target components.   • Determination of hemagglutination activity: We provide enzymatic animal erythrocytes BBL agglutination services. Add PBS, shake well, and leave at room temperature for some time before detection after serial dilution with the U-type microtiter plate.
  • Carbohydrate specificity analysis service: Different types of carbohydrates and their derivatives are serially diluted in the buffer. Observe agglutination after adding purified BBL and enzyme-enriched erythrocyte suspension.
  • Molecular weight determination: We use acrylamide gels for sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) determination and molecular weight determination. The molecular weight of the Native Lectin is determined by size-exclusion chromatography of the gel column.
  • Chemical analysis: Determination of carbohydrate content of purified lectins by phenol-sulfuric acid reaction.

Results: The lectin purified from the B. bubalis gave a single polypeptide band of 14.5 kDa corresponding to it under SDS-PAGE reducing conditions. Suggesting that it was composed of two identical noncovalent-linked 14.5 kDa subunits. The structure of the dimer was further confirmed by the Scatchard analysis, indicating the presence of two carbohydrate-binding sited for each native lectin molecule. It was also shown that the oxidized form was completely inactive, thus demonstrating that oxidative functional lectins in neural tissue may simply act as an autocrine or paracrine factor, and function more like a cytokine or chemokine.
