Ion Exchange Chromatography-based GSL Purification Service

Glycosphingolipids (GSLs) are minor components of cell membranes and are associated with cell growth, differentiation, and viral and oncogenic transformation. Extracted GSLs are usually mixtures with other polysaccharides. CD BioGlyco offers separation and purification services for GSLs based on ion exchange chromatography (IEC).

How Does IEC Work?

IEC is a chromatographic method that uses ion exchange resin or chemically bonded ion exchange agent as the stationary phase to achieve separation by using the difference in ion exchange capacity or selectivity coefficient of the separated components. The principle of IEC is that the charge of the separated substance can be combined with the opposite charge of the ion exchanger, and the combination between this charged molecule and the stationary phase is reversible. When changing the pH or eluting with a buffer that gradually increases the ionic strength, the substance bound on the ion exchanger exchange with the ions in the eluent and is eluted into the solution. Since different substances have different charges, their binding ability to the ion exchanger is also different, so the order of being eluted into the solution is also different, and thus they are separated.

Our Service

IEC is an effective technique for the separation and purification of GSLs. However, GSLs are complex molecules and other techniques may need to be used in their purification process.   • We combined cellulose column chromatography with IEC to achieve efficient separation of GSLs after removing non-polar substances that do not contain GSLs.
  • We integrated gel filtration with IEC to achieve the separation and purification of GSLs while removing the larger molecular weight aggregates of GSLs. Our service is adapted to complex mixtures containing GSLs, thus allowing scientists to determine the identification and concentration of each component and to separate GSLs components from other contaminating molecules for further analysis or application.
