Introduction

Human milk contains lacto-N-biose (LNB), also called lacto-N-biose I, which is a component of HMO and one of the Bifidus growth factors. Moreover, lacto-N-biocides could transform LNB into lacto-N-tetraose and lacto-N-neotetraose, the most prevalent constituents and key core structures of HMOs. Several cell types, such as the intestinal epithelium and blood group antigens, include LNB in the glycan domains of glycolipids and glycoproteins.

Analysis Methods

The most used quantitative analytical technique for HMOs especially LNB measurement is high-performance liquid chromatography (HPLC). Based on HPLC, it may be combined with other different detection methods, including mass spectrometry (MS), fluorescence, evaporative light-scattering (ELS), pulsed amperometric detection (PAD), and ultraviolet (UV). The most highly developed method for measuring HMOs which does not require derivatization is high-performance anion-exchange chromatography (HPAEC) with PAD. HMOs should be derivatized to produce the detection signal or boost the detection sensitivity by HPLC analysis when utilizing UV or a fluorescence detector due to the poor UV absorption and lack of fluorescent characteristics. For the analysis of HMOs, NMR and capillary electrophoresis (CE) is also employed.
