Introduction

Lectins are a diverse group of glycan-binding proteins in organisms. It has the characteristics of reversible binding to glycan chains, no modification of polysaccharides, and no enzyme catalytic activity. They have specific recognition capability for glycans and are often used as a "decoder" to elucidate the detailed structure information of glycan chains on carbohydrate conjugates. Lectins have been extensively used in the field of biochemistry, mainly for the detection and enrichment of glycans and glycoconjugates. The low affinity and broad specificity of lectins are considered to be suitable for "profiling" tasks to cover a wider range of ligands.

Lectin-based glycomics is a new comprehensive technology in post-genomic science. In recent years, CD BioGlyco has developed various lectin-based techniques such as lectin blots, lectin histochemistry/cytochemistry, lectin-antibody sandwich enzyme-linked immunosorbent assay (ELISA), lectin affinity chromatography, lectin microarray, and lectin microfluidics. Services

A variety of lectin-based techniques at CD BioGlyco have been successfully applied to glycoprotein analysis with high sensitivity. Services we provide include but are not limited to:

  • Lectin blot is used to detect glycoproteins separated by gel electrophoresis (1D-SDS-PAGE or 2D-SDS-PAGE) and transferred onto membranes for lectin blotting.
  • Lectin affinity chromatography was used as a way for targeting particular sub-glycoproteomes from clinical samples for the discovery of cancer-associated biomarkers.
  • We use lectin histochemistry/cytochemistry to directly observe the localization and distribution of glycans on glycoproteins in tissues and cells.
  • We have developed a new platform represented by a lectin microarray that can perform multiple glycan-lectin interaction analyses. These techniques provide customers with useful clues about the characteristic features of glycoconjugates including differences in terminal modifications (e.g., sialic acid bond, fucosylation type) and differences in higher ordered structures in glycan density, depth, and direction composed for N- and O-glycans.
