Monosialoganglioside 1(GM1) Analysis Service

Monosialoganglioside 1(GM1) Analysis Service

CD BioGlyco has extensive experience and a good reputation in the field of Lipidomics Analysis Services. We provide sensitive, reliable, and accurate Sphingolipid Analysis Services to solve customers' problems. We offer advanced liquid chromatography–electrospray ionization-mass spectrometry (LC-ESI-MS) method to quantify GM1 in samples.

Overview of Monosialoganglioside 1 (GM1)

GM1 is a major ganglioside subtype. They are involved in cell proliferation, differentiation, adhesion, signal transduction, cell-cell interactions, tumorigenesis, and metastasis. Accumulation of gangliosides has been linked to several diseases, including Tay-Sachs and Sandhoff disease. Furthermore, GM1 is abundantly present in the outer lobe of the plasma membrane of central nervous system neurons. They exhibit neuroprotective properties in the central nervous system, with GM1 protecting against lead-induced impairment of synaptic activity and oxidative damage to the hippocampus in vivo.

GM1 Analysis Service at CD BioGlyco

This service provided by CD BioGlyco is used to analyze Monosialoganglioside (GM). We offer advanced LC-ESI-MS method to quantify GM1 in samples. The method utilizes chemical derivatization techniques to increase the sensitivity for the determination of GM1 in samples. The method uses reversed-phase ultra-performance liquid chromatography (UPLC) for chromatographic separation and tandem mass spectrometry in multiple reaction monitoring (MRM) modes to enhance detection, sensitivity, and specificity. Taking plasma samples as an example, our analysis process is as follows.

  • Sample preparation

Preparing working plasma samples using plasma and internal standard working solution. After cooling and vortexing, the mixture is centrifuged and the supernatant is transferred to an autosampler vial with a micro-insert for LC/MS analysis.

  • LC-MS/MS analysis

After chemical derivatization, the sample is injected into a UPLC column. Elution and analysis are carried out according to a certain analysis procedure. The introduction of an electrospray ionization source is required during the analysis because all derivatized monosialoganglioside components are eluted and detected sequentially.

  • Calibration curve

Ganglioside quantification is performed on plasma samples using a calibration curve based on seven levels. For each of the three monosialogangioside species, the total peak area is calculated by summing the IS-normalized peak areas of the individual components based on a single injection.
