Background

Polysialic acids (polySia) are linear carbohydrate polymers which comprised of negatively-charged 5-N-acetylneuraminic acid (Sia, Neu5Ac) residues and connected by α-2,8-, α-2,9- glycosidic bonds or α-2,8 / α-2,9 alternating glycosidic bonds. In addition to polySia containing Neu5Ac units, other Sia units are also found in polySia polymers in nature: 5-N-hydroxymethylneuraminic acid (Neu5Gc) and deaminated- neuraminic acid (KDN). Polysialic acid regulates nerve cell development, nerve guidance and synapse formation by changing the adhesion of nerve adhesion molecules in the nervous system and also affects cell growth by interacting with cells and growth factors. The abnormal expression of polySia in cancer is highly correlated with disease progression. PolySia is also expressed on the surface of bacteria such as E. coli and is related to bacterial virulence. Therefore, quantifying the expression levels of polySia and polysialylated protein between normal and diseased tissues/cells is very important.

  • Fluorometric HPLC quantitative analysis: Fluorescence HPLC analysis has been proved to be very suitable for measuring the degree of polymerization of polySia. According to the final analyte, fluorescence quantitative analysis can be divided into two categories:
    • Direct method: quantifies polySia directly. HPAEC-FD, HPAEC-CAD
    • Indirect method: quantifies the derived hydrolysate by Fractionation-based RPLC-FD and Periodate fluorometric C7/C9 (sialic acid) analysis of polySia.
  • Quantitative immunodetection: we provide antibodies to detect and quantify polySia and polysialylated proteins in biological matrices by enzyme-linked immunosorbent assay (ELISA), Western blotting and flow cytometry.
