Solanum tuberosum Lectin (STL) Production Service at CD BioGlyco

STL is a glycoprotein isolated from potatoes. It contains 50% carbohydrates that are composed of arabinose and galactose. It consists of two subunits (50 kDa). To increase lectin production, the first step is to increase the amount of crude lectin extracted from the starting material. This is followed by the critical purification procedure. Extraction and purification

We have abundant isolation methods to extract STL from Solanum tuberosum tubers. There are various types of lectin extraction methods, including traditional precipitation methods, ultrasonic extraction, microwave extraction and so on. We will develop a STL production program according to the client's requirements for lectins. Traditional precipitation methods and emerging technologies are used in combination.

Lectins have carbohydrate-binding specificity. We have developed a variety of purification methods to meet customer requirements for lectin purity, including Gel ChromatographyAffinity Chromatography, and others.

Characterization

  • Bradford assay: To determine the protein concentration of the extracts of the lectin.
  • Haemagglutination assay: To detect the haemagglutination activity.
  • SDS-PAGE, size-exclusion chromatography and MALDI-TOF-MS: To determine the molecular weight of the fractions.

Results: In this study, a method was developed for the efficient extraction of heat-resistant lectins from potato tubers. The method involved ammonium sulfate precipitation, heat treatment, and ion exchange chromatography. The lectins extracted by the method were of high purity and the extraction time was greatly reduced.

Applications

  • STL is used for Lectin Microarray. Lectin microarrays provide a fast and sensitive method for the analysis of glycans and glycoproteins. Researchers have used lectin microarrays to analyze the glycoforms and glycosylation properties of glycoproteins.
  • STL is suitable for use in FLISA.
  • STL is used to visualize the localization of specific saccharide moieties and the distribution of plasma membrane glycoproteins.
