Analytical Characterization of Cyclodextrins

Cyclodextrins are cyclic oligosaccharides whose physicochemical properties can be significantly modified by forming clathrates with plenty of hydrophobic guest molecules. Both analytical chemists and pharmaceutical scientists have long been exploiting these properties for solubility enhancement of insoluble drugs, stability enhancement, protection against oxidation, light-induced reaction and loss by evaporation, etc. Cyclodextrins have no chromophores or fluorophores, so they cannot be detected using traditional HPLC detectors, such as fluorescence detectors. It's also challenging to detect cyclodextrins by high-performance liquid chromatography (HPLC). However, other HPLC detections such as mass spectrometric detector, evaporative light scattering detector (ELSD), refractive index (RI) detector, and charged aerosol detector (CAD) can also be applied. Today, cyclodextrins are considered to be versatile additives of many kinds of molecular recognition-based analytical methods, such as separation science, spectroscopic methods, electroanalyses, and single-molecule sensing.
