Analytical Characterization of Cyclodextrins
Cyclodextrins are cyclic oligosaccharides whose physicochemical properties can be significantly modified by forming clathrates with plenty of hydrophobic guest molecules. Both analytical chemists and pharmaceutical scientists have long been exploiting these properties for solubility enhancement of insoluble drugs, stability enhancement, protection against oxidation, light-induced reaction and loss by evaporation, etc. Cyclodextrins have no chromophores or fluorophores, so they cannot be detected using traditional HPLC detectors, such as fluorescence detectors. It's also challenging to detect cyclodextrins by high-performance liquid chromatography (HPLC). However, other HPLC detections such as mass spectrometric detector, evaporative light scattering detector (ELSD), refractive index (RI) detector, and charged aerosol detector (CAD) can also be applied. Today, cyclodextrins are considered to be versatile additives of many kinds of molecular recognition-based analytical methods, such as separation science, spectroscopic methods, electroanalyses, and single-molecule sensing. The development of novel and improved analytical technologies is highly valuable in the pharmaceutical field in order to better predict and understand the aggregation phenomenon as well as their detection and quantification.

Results: This review details the unique truncated cone structure of native cyclodextrins (α-, β-, and γ-cyclodextrin), characterized by a hydrophobic internal cavity and hydrophilic exterior, which underpins their ability to form inclusion complexes with various molecules. It explores key physicochemical properties like conformational flexibility, aqueous solubility variations, and aggregation behavior, emphasizing how chemical modifications (e.g., hydroxypropyl derivatives) enhance functionality. The article examines factors influencing complexation (cavity size, guest properties, water displacement) and discusses the broad applications of cyclodextrins, including pharmaceutical drug delivery (solubility enhancement, stability, bioavailability), environmental remediation (pollutant removal), food technology (flavor encapsulation, cholesterol reduction), textiles, cosmetics, and emerging uses in solar energy materials and sensors.
