  • SBA production service

We mainly extract lectins from soybeans or roots. The crude extract of SBA is usually prepared by ammonium sulfate step-by-step precipitation. It is cumbersome, and ammonium sulfate will cause a small part of protein denaturation. Therefore, it is essential to find an efficient and safe method to prepare the crude extract of SBA for quantitative purification. We have improved the method by referring to the reports on the separation of SBA. The optimized preparation method shortens the experimental period. It is verified that the SBA yield is improved.

Based on the fact that SBA specifically and reversibly binds to the corresponding ligands or ligands, we further improve its purification method. At present, we mainly use Affinity Chromatography for the preparation of SBA. We also try to use new purification methods to shorten the purification steps and simplify the experimental operation.

We also provide comprehensive SBA analysis services. We use polyacrylamide gel electrophoresis (SDS-PAGE) to identify the subunit composition of SBA and its molecular weight. SBA is found to bind specifically to GalNAc, galactose, and LacNAc. We will also test its amino acid composition, sugar-binding specificity, etc.

Results: In this study, a new method for the purification of lectin, called CaRe, was developed using SBA, etc. In this method, lectin was captured by a trapping agent (TCA). The captured lectin was released by competitive monovalent ligands. The purified lectin was later obtained by membrane filtration separation. The validation results of the separation of SBA, etc., showed that this method was accurate and simpler to operate than traditional protein purification methods such as ion exchange chromatography.
