Structure of Lacto-N-tetraose (LNT)

HMOs are a group of naturally occurring oligosaccharides (OS) present in human milk. They are composed of five basic units, including an acidic monosaccharide, sialic acid (Sia); an amino sugar, N-acetylglucosamine (GlcNAc); and three monosaccharides, L-fucose (Fuc), D-galactose (Gal) and D-glucose (Glc). It has been reported that about 200 different HMOs have been characterized. LNT is the most abundant HMO in breast milk and is a representative of neutral HMO. It is a tetrasaccharide composed of Gal, GlcNAc, and lactose connected sequentially via β1-3 bonds, Galβ1-3GlcNAcβ1-3Galβ1-4Glc. Most HMOs are derived from sialylation or fucosylation based on lacto-N-triose (GlcNAcβ1-3Galβ1-4Glc) and LNT.

Biological Significance of LNT

Nitrogen-containing LNT is considered one of the most important prebiotics in breast milk, supporting the growth of healthy bacteria in the gut microbiota, thereby promoting the development of the natural microbiota in newborns. LNT is one of the first functional foods that babies eat. LNT has been reported to be a non-competitive food source for Bifidobacterium infantis. Bifidobacteria not only cleave chemical bonds of LNT to digest OS in breast milk through specific transporters and glycosidases but also may inhibit the adhesion of harmful bacteria and the attachment of harmful bacteria proteins in the intestinal tract. In addition, LNT may also benefit the immune system in a supportive role.
