What Is 2'-Fucosyllactose (2'FL)?

2'FL is one of the key functional human milk oligosaccharides (HMOs) naturally present in human breast milk, accounting for about 30% of all HMOs. 2'FL is an oligosaccharide, more precisely, fucosylated, neutral trisaccharide composed of L-fucose, d-galactose, and d-glucose units via α-(1, 2)-linkage. It was first discovered in the 1950s in human milk. To date, 2'FL, with irreplaceable physiological effects, is one of the most hopeful Oligosaccharides in HMOs and has gathered great interest not only in large-scale production but also in commercial applications.

Applications of 2'FL

As the main HMO constituent, 2'FL has been considered to be a promising infant formula additive as well as in food due to its various beneficial effects on infant health. It is permitted to not only serve as the food ingredient added to infant formulas but also as dietary supplements and medical foods. Intensive efforts have thus been made to develop a synthetic method for the economic production of 2'FL on a large scale. So far, three approaches have been applied to produce 2'FL, including chemical, chemo-enzymatical, and cell factory methods.

Our Services

We use a HILIC-based method for the profiling and structural characterization of 2'FL. Oligosaccharides are fluorescently labeled by 2-aminobenzamide. Besides, this method contains the pre-fractionation of the derivatized compounds (in order to separate neutral and acidic species) by weak anion-exchange chromatography before subsequent separation using hydrophilic interaction chromatography. It is necessary to further purify before HPLC analysis, as the lactose in high concentration deteriorates the quality of the chromatograms. Structural characterization of the separated oligosaccharides is performed offline tandem mass spectrometry in negative ion mode following exoglycosidase digestion.
