Introduction of 6'SLN

The influenza virus hemagglutinin glycoprotein binds to Neu5Ac-2-6-Gal-1-4-GlcNAc (6'-sialyl-LacNAc; 6'SLN) on the surface of host cells, starting viral infections. All human influenza A and B viruses share the high-affinity receptor 6'SLN, therefore polymeric inhibitors that include this molecule may provide defense against all human pandemic strains. Furthermore, it is presently thought that for the virus to propagate effectively from person to person, 6'SLN-terminated receptors must be recognized by the virus. If this theory is correct, 6'SLN-based medications may offer excellent defense against newly developing pandemic influenza virus strains. High molecular weight conjugates of Neu5Ac2-3Gal1-4Glc (3'SL), Neu5Ac2-6Gal1-4Glc (6'SL), and 6'SLN were produced by connecting spaced oligosaccharides to polyacrylamide (N-oxi-succinimidyl acrylate).

Applications of 6'SLN

The interaction of the viral haemagglutinin (HA) with cell surface glycans mediates the earliest step of host cell infection by influenza A viruses (IAV). It is widely known that IAVs must bind to Gal(1,4)Glc/GlcNAc (lactose/lactosamine) glycans with a terminal (human receptors) or (avian receptors) N-acetylneuraminic residue that is often present on N-glycans. Unfortunately, little is known about the function and importance of sialylated Gal(1,3)GalNAc epitopes, which are typical O-glycoforms, in the pathogenesis of influenza viruses. Whereas 3'SLN is chosen by avian influenza strains as a key attachment site, 6'SLN is known to be exclusive to human influenza strains. Specifically binding to the hemagglutinin (HA) trimers of influenza strains are 6'SLN and 3'SLN.
