6'-Sialyllactose (6'SL), Abundant Sialylated Oligosaccharide

The 6'SL is composed of the monomers glucose, galactose, and N-acetylneuraminic acid. It is one of the most abundant sialylated (acidic) Oligosaccharides in the oligosaccharide fraction of human milk oligosaccharides (HMOs). 6'SL is structurally represented as Neu5Acα2, 6Galβ1, 4Glc. It is the simplest sialylated HMO with sialyl linked to lactose by an α2,3-glycoside bond. Sialylated HMOs account for approximately 13% of the total HMOs in molar ratio, in which 6'SL are present at approximately 6%. The concentrations of 6'SL change slightly during lactation. The mean concentrations of 6'SL are 0.40, 0.71, 0.40, and 0.30 g/L, at colostrum (0-5 d), transitional (6-14 d), mature milk (15-90 d), and late milk (>90 d).

Effects of 6'SL

6'SL have promising prebiotic effects, especially in promoting the proliferation of bifidobacteria and shaping the intestinal microbiota. It also displays a lot of other health effects, including antiadhesive antimicrobial ability, antiviral activity, prevention of necrotizing enterocolitis (NEC), immunomodulatory activity, regulation of intestinal epithelial cell response, promotion of brain development, and cognition improvement. The safety of 6'SL has been diffusely studied in piglets, rats, and healthy humans. These results suggest that 6'SL is safe and well-tolerant without toxic or genotoxic effects. It represents one of the most important sialylated HMOs and has been commercially added to infant formulas, generally in combination with neutral fucosylated HMOs, including 2'-fucosyllactose (2'FL) and 3-fucosyllactose (3FL). An increasing number of studies have focused on the exploration and verification of various physiological properties of HMOs. The physiological functions of 6'SL, one most important sialylated HMOs, are gaining increasing attention.

Our Services

CD BioGlyco has developed accurate, simple, and rapid methods for quantifying and analyzing 6'SL. We use capillary electrophoresis (CE) and CE-MS coupling which have been demonstrated to be an interesting technique for the analysis of 6'SL, requiring only simple sample preparation, safe reagents, and the use of a regular UV detector. CE and CE-MS are other state-of-the-art methods, which recently show a tremendous development. They are introduced to a broad range of research fields and are outstanding in view of separation efficiency and selectivity. Regarding the analysis of oligosaccharide mixtures, CE is well known for its high resolving power and sensitivity. Some commercially available HMO isomeric standards have even been successfully separated by CE, e.g. 2'fucosyllactose (2'FL)/3-sialyllactose (3'FL) or lacto-N-fucopentaose I/II (LNFP I/II).
