Amaranthus caudatus Lectin Production Service

Amaranthus caudatus Lectin Production Service

Amaranthus caudatus Lectin (ACL) Production Service at CD BioGlyco

ACL is extracted and purified from the A. caudatus plant, which is a dimeric glycoprotein composed of two subunits with a molecular weight between 60,000 - 70,000 and binds best under slightly acidic conditions. The isoelectric focusing of the lectin resolves it into four bands with isoelectric points between pH 6.7 and pH 7.7. CD BioGlyco has established a one-stop Plant Lectin Production solution, and we provide mature ACL production services.

  • Extraction
    • We extract ACL from A. caudatus plant tissue, usually using buffers or solvents to extract and break the cell wall to release lectins through mechanical disruption and stirring.
    • We use techniques such as centrifugation to spin the suspension to remove solid particles and obtain a clear extract.
    • We enrich ACL from the extract using different enrichment techniques such as salting out, gel filtration chromatography, or affinity chromatography.
  • Purification

We use further chromatography techniques (such as ion exchange chromatography, gel filtration chromatography, etc.) to purify the enriched lectin, remove other impurities, and obtain high-purity ACL.

  • Activity assessment

ACL is a protein with lectin activity that specifically binds to Galβ(1-3)GalNAc (T antigen). We perform activity assays on purified ACL samples using appropriate experimental methods and kits. Typically, the activity of a lectin is assessed by the agglutination of red blood cells caused by its binding to specific sugar residues. ACL is not specific for blood types, but has a higher affinity for type O red blood cells than type B and type A red blood cells.

  • Custom production

We provide different specifications and scale ACL production services. We aliquot the purified ACL into appropriate containers and store it as required to ensure its stability and shelf life. We offer several types of Products with ACL Conjugate that are used in glycobiology, immunohistochemistry, immunoassays, and western blotting.

Results: The authors used lectin microarrays to analyze endometrial cancer tissue and cell lines from patients and divided patients into different clinical stages based on cancer progression (Figure 2A). The results showed that there were significant differences in the relative binding strength of ACL to endometrial cancer in different stages, with the ACL signal in stage III/IV tissue being higher than that in stage I tissue (Figure 2B). The authors believed that the strong correlation between lectins and endometrial cancer stage provided a new way to determine treatment options and predict prognosis.

Applications

  • Glycoprotein research: ACL is used to study the structure and functions of glycoproteins, including analysis of glycosylation markers and glycosylation patterns, to gain an in-depth understanding of the biological properties of glycoproteins.
  • Cell surface sugar detection: ACL binds to specific sugar residues on the cell surface and is used as a detection tool for cell surface sugars. It is used for the identification of cell phenotypes, cell sorting, and the study of glycosylation heterogeneity.
  • Tumor marker research: ACL specifically binds to certain tumor markers and is an important tool in the detection and research of tumor markers, which is used for early tumor monitoring and research.

Advantages

  • CD BioGlyco has established a diversified Lectin Production solution, and our scientists are proficient in ACL extraction and purification technology to ensure the production of high-quality ACL.
  • CD BioGlyco has established strict quality control standards and procedures to ensure that the ACL produced meets expected purity, stability, and activity requirements.
