This highly responsive formulation features antisense oligonucleotide pH-sensitive proliposomes designed as a dry-state precursor for a stimulus-responsive nucleic acid complexation model. The solid microarchitecture consists of a water-soluble carbohydrate carrier network uniformly coated with dehydrated antisense oligonucleotides (ASODNs) alongside pH-sensitive lipids (such as dioleoylphosphatidylethanolamine and cholesteryl hemisuccinate). This dry configuration completely prevents the rapid chemical hydrolysis of the phosphodiester bonds and eliminates premature vesicle aggregation during extended storage. Upon standard hydration with an aqueous buffer, the matrix self-assembles into a uniform suspension of pH-sensitive liposomes that undergo phase changes under acidic conditions.
