The biological activity of PNA plays an important role in cell recognition, inflammatory response, blood clot formation, etc., which interacts with sugar molecules on the cell surface and participates in a variety of physiological and pathological processes. PNA has been widely used as a tumor marker because it specifically binds to the β-D-Gal(1-3)-D-GalNAc residue of the Thomson-Friedenreich antigen.   • PNA extraction and purification service
    • Grinding: PNA is a plant-derived protein, and we select healthy and high-quality peanut seeds for grinding.
    • Extraction: We mix peanut powder with an appropriate buffer solution such as phosphate buffer or tris buffer to extract PNA. Stirring or shaking the mixture at a low temperature for a while enhances the extraction effect.
    • Centrifugation and filtration: We centrifuge the extract to remove suspended solid particles and impurities, and then further filter the extract using filter paper or microporous filters to remove fine particles.
    • Precipitate formation and separation: We add a suitable precipitant (such as ammonium sulfate or polyethylene glycol) to the extraction solution to form a PNA precipitate, and then separate the PNA from the extraction solution by centrifugation.
    • Concentration: We redissolve the PNA precipitate in an appropriate buffer solution, and then use concentration techniques to concentrate the PNA in the solution.
    • Column chromatography: We use column chromatography technology to further purify the concentrated PNA solution and remove other impurities, such as ion exchange chromatography, gel filtration chromatography, or affinity chromatography.

PNA mainly exerts its biological activity by interacting with specific sugar molecules on the sugar chain, which binds with high affinity to the β-D-galactose structure on the cell surface or in solution. In addition, PNA has an agglutination effect, which brings red blood cells or other cells together to form agglutinates. Results: The authors constructed a label-free biosensor using PNA for the detection of T antigen, monitoring the formation of complexes between PNA and T antigen in glycoproteins by electrode impedance. The PNA biosensor was used to analyze serum samples from various cancer patients, the results showed that T antigen was highly expressed in skin cancer and breast cancer, and low in kidney cancer, liver cancer, and rectal cancer. The biosensor response was highly selective.
