Description

This highly advanced composite material features doxorubicin and small interfering RNA co-loaded lipid-enveloped mesoporous silica nanoparticles (CLMSNs-SS-NH2@DOX/siRNA) designed with an organic-inorganic hybrid core-shell architecture. The internal core structure consists of amino-functionalized, disulfide-linked mesoporous silica nanoparticles (CLMSNs-SS-NH2) that co-adsorb hydrophilic doxorubicin (DOX) hydrochloride and small interfering RNA (siRNA) fragments within their porous matrix, which is completely enclosed by a self-assembled outer shell of a phospholipid bilayer matrix. This complex configuration combines the rigid loading capacity and reduction-responsive cleavage parameters of a modified silica core with the interfacial stability and retention properties of a lipid bilayer shell. The resulting hybrid nanoparticles exhibit exceptional physical stability against cargo leakage and gravitational sedimentation.
