Description

This non-liposomal engineered reference formulation features doxorubicin loaded pH-responsive polymer micelles. The sub-micron architecture consists of a self-assembled core-shell amphiphilic block copolymer matrix (such as poly(ethylene glycol)-b-poly(poly(beta-amino ester)) or poly(acrylic acid)) that encapsulates the hydrophobic or base form of the anthracycline doxorubicin entirely within its hydrophobic block core. Under neutral pH conditions, the micellar matrix remains highly stable; however, exposure to mildly acidic buffers triggers immediate protonation of the responsive blocks, causing rapid micellar disassembly, core phase separation, and instantaneous cargo release.
