High-performance Liquid Chromatography (HPLC) for Glycan Profiling

High-performance liquid chromatography (HPLC) has been widely used as a sensitive technique for monitoring protein glycosylation. CD BioGlyco is committed to providing a wide range of glycan profiling services through HPLC methods for customers.

Background

HPLC, as a technique for the separation and analysis of glycoproteins and their derived carbohydrates, has gained significant interest in recent years. HPLC is an important branch of chromatography. With liquid as the mobile phase and high-pressure infusion system, the mobile phases such as single solvent with different polarity, mixed solvents with different proportions, and buffer are pumped into the chromatographic column equipped with the stationary phase. After being separated in the column, components enter the detector for detection to realize the analysis of samples.

According to the characteristics of glycans and the HPLC matrix used, the released glycans can be separated into multiple components. Normal phase and reverse phase chromatography are often used in combination with fluorescent-labeled oligosaccharides to separate sialylated and neutral N- and O-linked glycans. In addition, a method of high pH anion exchange chromatography with pulsed amperometric detection (HPAE-PAD) has been successfully used to separate and quantify the constituent monosaccharides released by acidic hydrolysis of glycan chains and to separate N-linked oligosaccharides separated by enzymatic digestion.

Analysis Strategies

CD BioGlyco has successfully developed a variety of advanced and sensitive strategies for glycoprotein analysis based on HPLC technology. The technologies we provide include but are not limited to:

  • A combination method of HPLC and fluorescent-labeled oligosaccharides for separation of sialylated and neutral glycans. The released glycans were labeled with fluorescence label, 4-aminobenzoic acid by reductive amination. Then the labeled glycans are separated by normal phase HPLC.
  • Separation of complex glycoprotein mixtures based on RP-HPLC followed by the analysis of the separated carbohydrates in combination with other detection techniques.
