The haemocytes are the source of serum lectin activity. The agglutination activity found in hemolymph serum has at least two components, lobster agglutinin I and II (LAg I and LAg II). LAg I binds mature T-thymocytes. Further explanation, LAg I is a sialic acid binding lectin that circulates as a multimer of 500-700 kDa, composed of 70 kDa subunits. LAg II has a specificity for N-acetyl-D-galactosamine residues (GalNAc).

  • Trypsin digests: Select samples are dialyzed against digestion buffer, concentrated by centrifugation, and processed for trypsin digestion. At the end of the reaction, detection is performed using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE).
  • Electron microscopy: When sufficient sample is available, purified lectin is examined by negative staining with phosphotungstic acid on a transmission electron microscope.
