In Vitro Anticancer Activity Screening Techniques

Currently, chemotherapy, surgery, radiotherapy, biotherapy, and other adjuvant therapy are common treatments for tumors. However, chemotherapy is the preferred and the most common treatment for patients in clinical practice. Yet, it still lacks effective antitumor drugs, while some patients easily acquire resistance to many new drugs after a period of treatment. Cancer cells are well known to display an enhanced sugar uptake and consumption. An increasing number of researchers from all over the world have been focusing on carbohydrate-based glycomedicine to find new antitumor drugs.

The MTT/MTS in vitro cell proliferation assay is one of the most widely used assays for evaluating the preliminary anticancer activity of glycomedicine. The highly reliable, colorimetric-based assay is readily performed on a wide range of cell lines. These are two of the most useful anticancer screening techniques available that are relatively economical and can be easily and routinely performed in the laboratory to characterize anticancer activity. Kinase inhibition assays are also one of the most widespread enzyme inhibition screening assays performed. Kinases are enzymes that play a key role in a number of physiological processes and their inhibitors have been found to exhibit anticancer activity against various human cancer cell lines.
