Description

This highly complex ternary gene-delivery model features liposome/polyethylenimine/antisense oligodeoxynucleotide complexes (Lipo/PEI/ASODN complexes). The structural framework utilizes a ternary lipoplex core architecture where single-stranded antisense oligodeoxynucleotide (ASODN) sequences are first electrostatically pre-condensed by the highly branched polycationic polymer polyethylenimine (PEI), and subsequently enveloped within a cationic or neutral helper-lipid liposome (Lipo) bilayer matrix. This advanced design introduces a highly robust positive surface charge profile and high mechanical stability from the polymer-lipid network, protecting the short oligodeoxynucleotide fragments from premature enzymatic cleavage or physical precipitation.
