The internal core structure consists of inorganic layered double hydroxide (LDH) synthetic clay nanoplates that intercalate the highly hydrophobic taxane paclitaxel (PTX) within their crystalline galleries, which is completely enclosed by a self-assembled outer shell of a high-purity phospholipid bilayer matrix. This complex configuration combines the rigid loading parameters of an inorganic host matrix with the biomimetic surface properties and interfacial stability of a lipid bilayer shell. The resulting nanohybrids exhibit exceptional physical stability against cargo leakage and gravitational sedimentation.
