Assay Principle
Nude mouse lipopolysaccharide (LPS) ELISA kit is designed to measure the level of nude mouse LPS in a sample. It utilizes purified nude mouse LPS antibody to coat the microtiter plate wells, creating a solid-phase antibody. The sample is then added to the wells, allowing the nude mouse LPS to bind to the solid-phase antibody. HRP-labeled antibodies specific to nude mouse LPS are subsequently added, forming an antibody-antigen-enzyme-antibody complex. After thorough washing, a TMB substrate solution is added, resulting in a blue color development due to the catalytic activity of the HRP enzyme. The reaction is stopped by adding a sulfuric acid solution, and the color change is measured at a wavelength of 450 nm using a spectrophotometer. The concentration of nude mouse LPS in the samples is determined by comparing the absorbance (O.D.) of the samples to a standard curve.
