  • Extraction and purification

PSL is present in the roots of peas, we use a series of processes such as crushing, dissolving, adjusting pH, centrifugation, and filtration to obtain a solution containing lectin. Next, the solution is concentrated and lyophilized to obtain the crude product.

Then, the crude product is purified and collected by Gel Filtration Chromatography, Ion Exchange Chromatography, Affinity Chromatography, and other purification methods. Finally, we get the high-purity PSL.

  • Characterization
    • Using sodium dodecyl sulfate-polyacrylamide gel electrophoresis method to determine the purity as well as the molecular weight of PSL.
    • Using Lowry's method, we have determined the protein content of PSL.
    • Use of the phenol-sulfuric acid method to determine the neutral sugar content of PSL.
    • Using atomic absorption spectrometry, to determine the metal ion content of PSL.
    • Using the hemagglutination assay and semi-antigen inhibition assay, the hemagglutination activity and sugar specificity are determined, respectively.

  • Results: The mannose/glucose-specific lectin was isolated and characterized from Pisum sativum. Firstly, it was purified and molecular weight (50 kDa) was determined using anion exchange chromatography, fast protein liquid chromatography, and size exclusion chromatography. The results showed that it was displayed as a heterotetramer, consisting of two distinct polypeptide bands with molecular weights of 6 kDa and 19 kDa, respectively. The terminal sequence of PSL showed some degree of homology compared to lectins from other legumes. Its hemagglutination activity was inhibited by glucose, mannose, and sucrose, and was attenuated at pH values above 12 or below 3. And it had good thermal stability. In addition, PSL showed mitogenic effects on murine splenocytes and inhibited HIV-1 reverse transcriptase activity.

Applications

  • PSL is used to isolate cells, separate glycoproteins, and glycopeptides, and differentiate normal cells from virus-transformed cells.
  • PSL is used to recognize glycoproteins, glycopeptides, and carbohydrates in biofilms and as a symbiotic medium for plants and microorganisms.
  • PSL lectin is used to research the biological properties of cells and the mechanism of action of glycoconjugates.
