Deoxyribonuclease I from Pichia pastoris is an endonuclease that plays a key role in setting up phosphodiester bonds, preferentially adjacent to pyrimidine nucleotides, yielding polynucleotides with a free hydroxyl group at the 3' position.

One unit causes an increase in absorbance at 260 nm of 0.001 per minute at 25 when acting upon highly polymerized DNA at pH 5.0.
